RESUMO

The chapter analyses the character of American politics and the significance of the transition from the presidency of Republican Donald Trump to Democrat Joe Biden. It assesses the impact of Trump's administration, including his refusal to recognize the legitimacy of the 2020 election result, on the increasingly polarized nature of party competition and on the divisions within American society. It evaluates the governing challenges facing the new administration which stem from its fragile control of Congress, bitter partisan opposition from the Republican Party and the range of serious policy problems to be addressed. Prominent among these policy problems (which are explored in detail in the subsequent chapters) are the management of the Covid-19 pandemic and the rebuilding of the American economy in its aftermath. The chapter uses the initiatives of Biden's first one hundred days in office to give a preliminary view of the new administration's approach. The chapter links the analysis of individual chapters on particular institutions and policy areas to the wider political framework and the functioning of American democracy. © The Author(s), under exclusive license to Springer Nature Switzerland AG 2022. As the ninth edition, this book reviews a time of political controversy in the United States, touching on topics such as gender, economic policy, gun control, immigration, the media, healthcare, the COVID-19 pandemic, and the widespread social protests against police brutality. The book looks both backwards to Trump's presidency and forward to Biden's. Using interviews with 15 Australian fathers, we explored the impact of having spent time in COVID-19 lockdown on men's views of their relationship with their children and family-work life balance. All interviewees were married to women and living with their children, most were employed and working full-time. Three themes were identified from the interviews: an ongoing desire to be present as a father, benefits to being present through COVID-19 lockdowns for self as a father and for the children, and conflicting pressures from workplaces and at home which were barriers to being present. Survey data 1 year later revealed that many of these fathers had shifted their work patterns because of their lockdown experience. Shifts in workplace culture and behavior were identified as supporting flexible working arrangements. Despite the immediate challenges of lockdown, it provided the opportunity for some fathers to reevaluate their priorities resulting in long-term changes in working patterns.

The arrangement of natural and physical features on the earth's surface are a few among the countless items that govern the airborne acoustic transmission at boundary layers. In particular, if the acoustic waves are attributes of live concerts at open-air theatres, without losing the sheen and quality, the audience should certainly receive the unbroken depth of the performance. Hence, at all times, it is advisable to analyse the auditory receptiveness, particularly in all intended recreational spaces. The current pandemic circumstances and the mandated COVID-19 prevention protocols encourage gatherings in naturally ventilated outdoor regions than confined indoors. This work predicts and quantifies the acoustic experience at the naturally carved amphitheatre at SAINTGITS, an autonomous institution at the down South-West of the Indian Subcontinent. The entire recreational space at SAINTGITS AMPHI was separately modelled as a Base case and Advanced case, and were analysed using the acoustic modelling module of EASE Focus, a renowned simulation freeware, which is in strict adherence with the International standards. The variation in loudness received at the nearest and farthest ends of the amphitheatre was between 67 to 80 dB. Though the Zero frequency SPL (Z-weighting) exhibited the loudness in the range of 81 to 85 dB and could maintain a safer auditory level for any human ear, it was confined to a hemispherical region near the sound source. A vertical beam angle of -4.0 degrees was found to be effective throughout. The procedures and analyses will certainly help the future organizers and stakeholders to effectively plan the resources to reap rich acoustic experience at terrain-centric locales. The surface topography and contours were plotted with another set of freeware, the CADMAPPER and the QUIKGRID, to compare terrain gradient with the known data. Furthermore, this interdisciplinary research exhibits the extensive simulation capability of both EASE Focus and QUIKGRID and demonstrates the modelling versatility and deliverable potential of these freeware to benefit the budding architects and researchers.

We introduce a target capture next-generation sequencing methodology, the ONETest Coronaviruses Plus, to sequence the SARS-CoV-2 genome and select loci of other respiratory viruses. We applied the ONETest on 70 respiratory samples (collected in Florida, USA between May and July, 2020), in which SARS-CoV-2 had been detected by a PCR assay. For 48 of the samples, we also applied the ARTIC protocol. Of the 70 ONETest libraries, 45 (64%) had a (near-)complete sequence (>29,000 bases and >90% covered by >9 reads). Of the 48 ARTIC libraries, 25 (52%) had a (near-)complete sequence. In 19 out of 25 (76%) samples in which both the ONETest and ARTIC yielded (near-)complete sequences, the lineages assigned were identical. As a target capture approach, the ONETest is less prone to loss of sequence coverage than amplicon approaches, and thus can provide complete genomic information more often to track and monitor SARS-CoV-2 variants.

The emergence of SARS-CoV-2 variants harboring mutations in the spike (S) protein has raised concern about potential immune escape. Here, we studied humoral and cellular immune responses to wild type SARS-CoV-2 and the B.1.1.7 and B.1.351 variants of concern in a cohort of 121 BNT162b2 mRNA-vaccinated health care workers (HCW). Twenty-three HCW recovered from mild COVID-19 disease and exhibited a recall response with high levels of SARS-CoV-2-specific functional antibodies and virus-specific T cells after a single vaccination. Specific immune responses were also detected in seronegative HCW after one vaccination, but a second dose was required to reach high levels of functional antibodies and cellular immune responses in all individuals. Vaccination-induced antibodies cross-neutralized the variants B.1.1.7 and B.1.351, but the neutralizing capacity and Fc-mediated functionality against B.1.351 was consistently 2- to 4-fold lower than to the homologous virus. In addition, peripheral blood mononuclear cells were stimulated with peptide pools spanning the mutated S regions of B.1.1.7 and B.1.351 to detect cross-reactivity of SARS-CoV-2-specific T cells with variants. Importantly, we observed no differences in CD4+ T-cell activation in response to variant antigens, indicating that the B.1.1.7 and B.1.351 S proteins do not escape T-cell-mediated immunity elicited by the wild type S protein. In conclusion, this study shows that some variants can partially escape humoral immunity induced by SARS-CoV-2 infection or BNT162b2 vaccination, but S-specific CD4+ T-cell activation is not affected by the mutations in the B.1.1.7 and B.1.351 variants.

Background Genomic sequencing is important to track and monitor genetic changes in SARS-CoV-2. We introduce a target capture next-generation sequencing methodology, the ONETest Coronaviruses Plus, to sequence SARS-CoV-2 genomes and select genes of other respiratory viruses simultaneously. Methods We applied the ONETest on 70 respiratory samples (collected in Florida, USA between May and July, 2020), in which SARS-CoV-2 had been detected by a qualitative PCR assay. For 48 (69%) of the samples, we also applied the ARTIC protocol for Illumina sequencing. All the libraries were sequenced as 2x150 nucleotide reads on an Illumina instrument. The ONETest data were analyzed using an in-house pipeline and the ARTIC data using a published pipeline to produce consensus SARS-CoV-2 genome sequences, to which lineages were assigned using pangolin. Results Of the 70 ONETest libraries, 45 (64%) had a complete or near-complete SARS-CoV-2 genome sequence (> 29,000 bases and with > 90% of its bases covered by at least 10 reads). Of the 48 ARTIC libraries, 25 (52%) had a complete or near-complete SARS-CoV-2 genome sequence. In 24 out of 34 (71%) samples in which both the ONETest and ARTIC sequences were complete or near-complete and in which lineage could be assigned to both the ONETest and ARTIC sequences, the SARS-CoV-2 lineage identified was the same. Conclusions The ONETest can be used to sequence the SARS-CoV-2 genomes in archived samples and thereby enable detection of circulating and emerging SARS-CoV-2 variants. Target capture approaches, such as the ONETest, are less prone to loss of sequence coverage probably due to amplicon dropouts encountered in amplicon approaches, such as ARTIC. With its added value of characterizing other major respiratory pathogens, although not assessed in this study, the ONETest can help to better understand the epidemiology of infectious respiratory disease in the post COVID-19 era.

COVID-19 has attracted global attention due to its rapid spread around the world with substantial morbidity and associated mortality. Severe COVID-19 can be complicated by the acute respiratory distress syndrome, sepsis and septic shock leading to death. These complications are thought to result from an overactivation of the immune system, leading to a cytokine storm syndrome associated with multiple organ failure. Here, we report that high mobility group box 1 (HMGB1), a prototypical damage-associated molecular pattern (DAMP) and a central mediator of lethal inflammation, could be a potential target for innovative therapeutic strategies for COVID-19. Serum HMGB1 in severe COVID-19 patients is elevated (189.40 ± 140.88 ng/ml). Exogenous HMGB1 induces the expression of SARS-CoV-2 entry receptor ACE2 in alveolar epithelial cells in an AGER-dependent manner. Importantly, genetic (using AGER siRNA) or pharmacological (using glycyrrhizin, chloroquine, hydroxychloroquine, and FPS-ZM1) inhibition of the HMGB1-AGER pathway blocks ACE2 expression. Thus, HMGB1 inhibitors are likewise promising drug candidates for the treatment of patients suffering from COVID-19.
